This study used different doses of hBMMSC to act on MCAO rats and examined the effects of hBMMSC under different concentration gradients and time conditions. The results showed that hBMMSC treatment could significantly improve the sensory and motor function of stroke rats and reduce the volume of cerebral infarction. In addition, different doses of hBMMSCs all reflect their therapeutic effects on brain rate. The low-dose group, injected with 1x106 cells, showed significant therapeutic effects in terms of neurobehavioral recovery and stroke reduction. The inhibitory effect of medium and high doses of hBMMSCs on the activation of small stem cells is stronger. This result suggests that hBMMSC may be an effective treatment for ischemic stroke.
The study findings reveal that hBMMSC treatment significantly ameliorates social deficits in BTBR mice, as evidenced by improvements observed in the Open Field Test (OFT), Light-Dark Box Test (LBT), Novel Object Recognition (NOR), and Free Social Test (FST), while also markedly reducing stereotypical behaviors. Furthermore, hBMMSC administration effectively reversed the alterations in microbial community composition in BTBR mice, particularly restoring the balance between Firmicutes and Bacteroidetes. Notably, several specific taxonomic groups correlated with ASD prognosis and behavioral scores, suggesting that intravenous injection of hBMMSC exerts beneficial effects on ASD by not only alleviating social impairments but also modulating gut microbiota composition. These findings highlight nBMMSC intravenous transplantation as a promising therapeutic strategy for potentially improving ASD symptoms.
